چکیده

T hrombosis is a dynamic process in which procoagulant activity, which promotes platelet activation and fibrin deposition, is balanced by physiologic anticoagulant and fibrinolytic activity. Historically, the treatment of thrombosis has been based on the use of agents that either inhibit procoagulant activity (i.e., anticoagulants) or induce fibrinolytic activity (i.e., plasminogen activators). However, recent data suggest that the success of antithrombotic interventions requires both inhibition of factors that promote thrombosis and induction of clot lysis. Thus, even when intense fibrinolytic activity is induced in patients with myocardial infarction, increases in procoagulant activity appear to be an important determinant of recurrent thrombosis.1-3 Results of recent clinical trials are consistent with the importance of conjunctive therapy with aspirin and heparin in patients given fibrinolytic agents.4-9 These

Thrombin as an anticoagulant.

Thrombosis is the most prevalent cause of fatal diseases in developed countries. An antithrombotic agent that can be administered to patients with severe acute thrombotic diseases without the risk of causing hemorrhage, as experienced with antithrombotic/thrombolytic therapy in the treatment of acute ischemic stroke or systemic anticoagulants like heparin, would likely revolutionize the treatme...

Effects of hormone replacement therapy on plasma and tissue fibrinolytic activity in a rat model of surgically induced menopause.

PURPOSE The purpose of this study was to analyze the effects of estrogen deficiency and hormone replacement therapy (HRT) on fibrinolytic activity in a rat mode of surgically-induced menopause. METHODS Twelve-week-old, sexually mature female Sprague-Dawley rats, each weighing 200-250 g, were randomly divided into four groups: (1) sham-operated group, (2) ovariectomy group, (3) ovariectomy gro...

Levels of three hemostatic factors in relation to serum lipids. Monocyte procoagulant activity, tissue plasminogen activator, and type-1 plasminogen activator inhibitor.

To explore the relationship between blood lipid levels and a predisposition to thrombosis, levels of three hemostatic factors were measured in 41 human subjects and correlated with serum lipids. Procoagulant activity associated with peripheral blood monocytes isolated and purified after a 2-hour incubation in whole blood was not significantly related to lipid levels. However, activity in monocy...

Polysulfated Trehalose as a Novel Anticoagulant Agent with Dual Mode of Action

Physiological hemostatic balance is a coordinated outcome of counteracting coagulation and fibrinolytic systems. An imbalance of procoagulant and anticoagulant factors may result in life threatening thromboembolism. Presently, anticoagulant administration is the first line of therapy for the treatment of these conditions and several anticoagulants have been approved, including various forms of ...

In-vitro and In-vivo Screening Methods for Antithrombotic Agents

Thrombolytic therapy and mechanical interventions are frequently used in the treatment of thrombotic diseases. The basic problems with current thrombolytic therapy include slow and incomplete thrombolysis and frequent bleeding complications. Therefore, there is a need of continuity to develop & screen out the various natural agents that possess antithrombotic activity by using different screeni...
